Crimson Publishers: Novel Approaches in Cancer Study

Cancer immunology largely depends on the effects of radiation therapy (RT). RT is predominantly focused on inducing tumor cell death, triggering anti-tumor immune responses, and generating DNA damage. Although the immune system has the ability to recognize and reject specific tumors, some tumors acquire characteristics that allow them to evade immune destruction by expressing a highly immunosuppressive microenvironment and decreasing their immunogenicity. Exposing the tumor site to radiation stimulates tumor-specific antigens, making them perceptible to the immune system and stimulating the priming and activation of cytotoxic T cells. In addition, tissue damage caused by radiation applied to the tumor microenvironment moderates a release of inflammatory cytokines that assists in leukocyte infiltration to the damage site and promotes an adaptive immune response. Furthermore, RT can reinforce tumor cell susceptibility to T cells, enhancing their eradication and apoptosis. Cancer cell apoptosis can manifest in the form of immunogenic cell death (ICD) or non-immunogenic cell death (non-ICD). While non-immunogenic cell death fails to elicit an immune response, immunogenic cell death specializes in antitumor immunity by modifying the surface composition of the cell and releasing soluble mediators, such as danger signals, to prompt elimination of the tumor cells. With advancement in understanding of immune cell types and pathways involved in the immune response, numerous preclinical studies have suggested that a combination of radiation and immunotherapy could lead to a propitious strategy against the challenges of cancer treatment. This paper examines the remedial adjustment of tumor immune responses and how it synchronizes efficiency and resilience to RT as well as its immunologic consequences.

Recent Advances in the Therapeutic Approaches of Glioblastoma Multiforme_Crimson Publishers

Recent Advances in the Therapeutic Approaches of Glioblastoma Multiforme by Siddharth Shah in  in Novel Approaches in Cancer Study Abstract Glioblastoma Multiforme (GBM, WHO grade IV) is one of the most aggressive, invasive, and lethal intracranial neoplasms, with a low post-diagnosis survival rate. Standard-of-care treatment regimens involving maximal surgical resection, radiotherapy, and genetic anti-tumor compounds like Temozolomide have only been marginally effective in improving overall survival and quality of life. Cell fusion, autophagy, and other complex biological processes affecting GBM pathophysiology are being studied to improve GBM treatment. This paper therefore focuses on oncolytic virus therapy combined with surgical resection, photodynamic therapy, and novel gene therapy, demonstrating how GBM treatment for patients could result in immediate and authentic tumor cytotoxicity and removal, rather than treatment of recurrent GBM. Standard therapy for GBM,...

Release of Inorganic Phosphate into the Tumor Environment: Possible Roles of Ecto- Nucleotidases and Ecto-Phosphatases

Abstract Inorganic phosphate (Pi) is essential for several biochemical reaction. Serum Pi is maintained at relatively narrow range concentrations, between 0.7 and 1.55mM. Tumor microenvironment presents a high Pi concentration (1.8±0.2mM Pi) and this could be associated wi the rapid growth in the “Growth Rate Hypothesis”. Several studies have identified high expression of Pi transporters in various tumor tissues. Similarly, ecto-enzymes (like ecto-nucleotidases or ecto- phosphates) act by dephosphorylating phosphosubstrates in the extracellular environment, and its high expression has been observed in various types of cancer. Little is known about the function of these ecto-enzymes on Pi releasing and accumulation in the tumor environment. Therefore, the purpose of this study is to correlate a possible contribution of the Pi release in the tumor microenvironment by ecto-nucleotidases and ecto-phosphatases, concomitant to the regulation of Pi extracellular pool by specific Pi...
